Product Comparison Data

1、Comparison of Tumor Formation Between Absin and Imported Brands

When using Absin's high-concentration matrix gel, the volume of subcutaneous tumors in mice was significantly higher than that of the control group and Brand C in the later stages.

2、Comparison of Organoid Culture Between Absin and Imported Brands

Using Absin's low-factor matrix gel, the number and volume of colorectal cancer organoids are significantly greater than those of a certain imported brand.

When using Absin's low-factor matrix gel, the number of colorectal cancer organoids is significantly higher than that of a comparable brand, and the morphology of the organoids stained with fluorescence is more intact. Human cholangiocyte organoids were imaged with staining by the nuclear dye DAPI (blue), the tight junction protein antibody anti-ZO1, and the cytoskeletal protein F-actin dye Alexa Fluor 647 Phalloidin.

Using Absin's low-factor matrix gel, the staining effect of hepatocellular carcinoma (HCC) organoids is comparable to that of a comparable brand. The HCC organoids were imaged with staining by the nuclear dye DAPI (blue), antibodies against the liver cell biomarker HNF4a and the HCC marker GPC3.

3、Comparison of Tube Formation Experiments Between Absin and Imported Brands

Both Absin's standard matrix gel and the matrix gel from a foreign competitor can support the formation of vascular networks by human umbilical vein endothelial cells (HUVEC).

Matrix Gel User Guide

1、After receiving the goods, please check the following:

1.1、Is there dry ice in the foam box?

1.2、Is the matrix gel solid and is the surface flat?

1.3、Is the color of the matrix gel within the normal range (white/milky white without phenol red, yellow-pink-red with phenol red, determined by temperature changes)?

1.4、If you do not plan to use the matrix gel immediately after receiving it, it should be stored in a -20/-80°C freezer for long-term preservation.

2、Before using the matrix gel each time, thawing operations should be carried out strictly according to the following matrix gel operation procedures!!!

2.1、Pre-cool the "Matrix Gel Ice Box (abs7289)" (or prepare crushed ice) and a 200μL pipette tip box for more than one hour at -20°C. Before leaving work in the evening, take the matrix gel out of the -20/-80°C freezer and place it in the "Matrix Gel Ice Box" (or bury the matrix gel in an ice box filled with crushed ice), then place the entire ice box in a 4°C refrigerator to thaw overnight.

2.2、The next day, after the matrix gel has thawed, it can be used. Take the matrix gel out of the 4°C refrigerator and immediately place it in the pre-cooled -20°C "Matrix Gel Aliquot Ice Box" (crushed ice box), then proceed with sterile use on a laminar flow hood/biosafety cabinet.

2.3、When using, take out the pre-cooled 200μL pipette tips from -20°C and open the matrix gel bottle cap (try to ensure that the matrix gel bottle is operated on the matrix gel thawing ice box).

2.5、After using the matrix gel, immediately put it back in the ice box and store it in a 4°C refrigerator for short-term preservation, and use it up within two weeks.

3、Essential precautions:

3.1、Do not directly touch the matrix gel with the palm of your hand;

3.2、The entire use process should be carried out on the pre-cooled matrix gel aliquot ice box (crushed ice box), and the remaining matrix gel from this experiment can be stored at 4°C for up to two weeks;

3.3Never thaw and store the matrix gel directly in the refrigerator, always place it in the "Matrix Gel Ice Box" (or bury the matrix gel in an ice box filled with crushed ice) before placing it in the 4°C refrigerator to thaw or store; in addition, do not thaw or store it on the edge of the refrigerator door, or in a refrigerator that is often opened to take out reagents, try to place it inside/drawer/cold air outlet of the 4°C refrigerator to thaw or store;

3.4Since the matrix gel is sensitive to temperature, it is recommended to use a non-defrost type and temperature-stable refrigerator for melting and storage, please do not thaw the matrix gel in a refrigerator with automatic defrost.

3.5If using a crushed ice box to thaw overnight, fix the position of the matrix gel bottle to ensure that the bottle will not tip over after the crushed ice melts overnight, and the ice water will not submerge the bottle mouth.

3.6The matrix gel should avoid repeated freezing and thawing during use.

3.7Liquid matrix gel will quickly gel at above 8°C (especially high-concentration matrix gel). Therefore, when using, the consumables that come into contact with the matrix gel must be pre-cooled (such as pipettes, tips, EP tubes, etc.), and the entire experiment must be carried out under sterile, ice conditions.

3.8The matrix gel should be in a liquid state on the ice when using. If the matrix gel has already solidified on the ice, it indicates that the aliquot operation did not maintain low temperature, causing the matrix gel to gel and should not be used again.

3.9If you find that the matrix gel in the 4°C refrigerator is very viscous but still liquid, you can place it in a pre-cooled -20°C ice box and let it sit in the -20°C refrigerator for 3-5 minutes, and the fluidity will generally return to normal; if it cannot be restored, it may be due to the refrigerator temperature being too high causing the gel, it is recommended not to use it.

4、Q&A

Q1 Why is my matrix gel very viscous?

A1 The higher the protein concentration of the matrix gel, the more viscous the gel. If the concentration is higher than 13.0mg/mL, the matrix gel will appear very thick. Matrix gel products are relatively viscous before dilution. The viscous, high-concentration matrix gel can be used directly without dilution, such as for culturing tumor cells and/or angiogenic factors, injected into mice, cells can remain in place, facilitating in situ analysis and/or subsequent removal; or diluted, used according to the standard concentration matrix gel product usage method, the specific dilution concentration is determined according to the experimental needs.

A2 In addition to the high concentration of the product itself causing viscosity, the state of the matrix gel is also related to temperature changes during transportation and storage conditions. Dry ice must be used throughout the transportation process. If the refrigerator where the matrix gel is stored has an automatic defrost function, the temperature rise during the defrost process may cause the matrix gel to gel. Therefore, never store the matrix gel in such a refrigerator with automatic defrost. To ensure the effectiveness of the matrix gel, the number of freeze-thaw cycles should be minimized as much as possible. Special attention: The temperature transfer of water, ice, ice water, and air is different, and air temperature transfer is particularly slow and unevenly distributed, so the matrix gel cannot be directly thawed in a 2-8°C refrigerator, it must be placed in an ice box/ice water mixture and then placed in a 0-4°C refrigerator to thaw.
