NK cells are lymphocytes crucial for innate immunity. They can lyse virus - infected, tumor, and abnormal cells without prior antigen stimulation. Constituting 5% - 15% of peripheral blood lymphocytes, NK cells are vital for immune surveillance.

Functionally, NK cells can be categorized into two subsets:
  1. Cytotoxic: Induce target - cell apoptosis via perforin and granzyme release.
  2. Immunomodulatory: Activate other immune cells by secreting cytokines like IFN - γ and TNF - α.
In humans:
  • CD56dimCD16bri NK cells: High cytotoxic activity.
  • CD56briCD16dim NK cells: High cytokine - secreting activity.
In mice:
  • CD11briCD27low NK cells: High cytotoxic activity.
  • CD11briCD27high NK cells: High cytokine - secreting activity.
When designing NK - cell detection panels, CD3 should be included to exclude T - cell interference, as NK and some T cells (e.g., NKT cells) share key NK - cell markers.
