MDSCs are derived from bone marrow progenitor cells and immature myeloid cells (IMCs). Normally, they are precursors to dendritic cells, macrophages, and granulocytes. They can rapidly differentiate into mature granulocytes, DCs, and macrophages, which then migrate to respective organs and tissues to perform normal immune functions. IMCs constitute approximately 0.5% of peripheral blood mononuclear cells. However, under pathological conditions like tumors, infections, inflammation, sepsis, and surgical trauma, the maturation of these myeloid - derived precursor cells is blocked due to cytokines. As a result, they remain at various differentiation stages and become MDSCs with immune - suppressive functions. Acting on cytokines, MDSCs are recruited, migrate, and expand. Their quantity and proportion in peripheral blood increase by about tenfold, accounting for roughly 10% of peripheral blood mononuclear cells in patients and spanning the entire disease - development process.
