IHC shows positive staining in paraffin-embedded E14.5 mouse embryo. Anti-Wnt3a antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Background
Wnt3a is a secreted, palmitoleated glycoprotein of ~39 kDa that acts as a principal ligand for canonical Wnt signaling; upon binding to Frizzled and LRP5/6 co-receptors it triggers a β-catenin–dependent cascade leading to TCF/LEF-mediated transcription of genes that govern embryonic axis formation, neural crest specification, self-renewal of hematopoietic and intestinal stem cells, and osteoblast differentiation, while its dysregulation drives colorectal, breast and hematologic malignancies, and recombinant Wnt3a is widely used in vitro to expand organoids and maintain pluripotency.
