Background

UBR2 is an evolutionarily conserved E3 ubiquitin ligase of the N-end rule pathway that selectively recognizes and ubiquitinates proteins bearing specific N-terminal destabilizing residues, thereby targeting them for proteasomal degradation; it contains an N-terminal UBR box that binds type-1 (basic) and type-2 (bulky hydrophobic) N-degrons, a central RING finger domain that coordinates ubiquitin-charged E2 enzymes, multiple auto-inhibitory and substrate-recognition motifs, and C-terminal regions that mediate interactions with transcription factors, chromatin remodelers, and DNA damage response proteins, enabling UBR2 to regulate diverse processes such as spermatogenesis, neurogenesis, cardiac development, genomic stability, and metabolic homeostasis through both proteolytic and non-proteolytic ubiquitylation events.

Immunocytochemistry

ICC shows positive staining in 293T cells. Anti-UBR2 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
