Background
TRIM63, also known as MuRF1 (Muscle RING Finger 1), is a muscle-specific E3 ubiquitin ligase encoded by the TRIM63 gene, primarily expressed in cardiac and skeletal muscle, where it plays a critical role in regulating muscle protein homeostasis by ubiquitinating sarcomeric proteins such as MYH6, MYBPC3, and cardiac troponin I for proteasomal degradation, thereby influencing muscle atrophy and hypertrophy; structurally, TRIM63 belongs to the tripartite motif (TRIM) family and contains an N-terminal RING domain, B-box, coiled-coil motifs, and a C-terminal acidic tail, and mutations in TRIM63 have been implicated in human hypertrophic cardiomyopathy due to impaired E3 ligase activity and disrupted protein degradation pathways.

IHC shows positive staining in paraffin-embedded human cardiac muscle. Anti-TRIM63 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
