Background

Transketolase (TKT) is a thiamine-diphosphate-dependent homodimeric enzyme encoded by the TKT gene on chromosome 3p21.1 that catalyzes reversible transfer of a two-carbon ketol group between ketose and aldose sugar phosphates, thereby linking the pentose phosphate pathway to glycolysis, generating NADPH and ribose-5-phosphate for nucleotide biosynthesis, and playing a central metabolic role in all organisms from bacteria to humans. Altered TKT activity—observed in diabetes and many cancers—makes it a potential therapeutic target, as its inhibition (e.g., by oxythiamine) suppresses cancer cell growth and thiamine supplementation can restore deficient activity .

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-TKT antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Negative control: IHC shows negative staining in paraffin-embedded human skeletal muscle. Anti-TKT antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in HepG2 cells. Anti-TKT antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
