Background
Terminal deoxynucleotidyl transferase (TdT), also called DNA nucleotidylexotransferase or terminal transferase, is a template-independent DNA polymerase encoded by the DNTT gene on human chromosome 10q23-q24 that is selectively expressed in immature lymphoid precursors (pre-B and pre-T cells) and certain leukemias/lymphomas; it catalyzes the non-templated addition of deoxynucleotides to the 3'-OH ends of DNA, generating N-region diversity at V(D)J junctions during immunoglobulin and T-cell receptor gene rearrangement, thereby expanding antigen-receptor repertoires critical for adaptive immunity, and can utilize a broad range of divalent cations (Mg²⁺, Mn²⁺, Zn²⁺, Co²⁺) as cofactors .

Immunocytochemistry
ICC shows positive staining in MOLT-4 cells. Anti- TdT antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
