Background

SOX10 (SRY-box transcription factor 10) is a 55-kDa member of the SOX family of high-mobility-group (HMG) DNA-binding transcription factors that acts as a master regulator of neural crest development, melanocyte specification, and oligodendrocyte differentiation by forming homodimers or heterodimers with other SOX proteins and binding to the consensus sequence A/TA/TCAAA/TG, thereby activating or repressing target genes such as MITF, MBP, and PLP1; mutations or chromosomal rearrangements affecting the SOX10 locus on 22q13.1 are linked to the neurocristopathy syndromes Waardenburg–Hirschsprung disease type 4 (WS4), PCWH (peripheral demyelinating neuropathy, central dysmyelinating leukodystrophy, Waardenburg syndrome, and Hirschsprung disease), and a subset of melanoma tumors where SOX10 serves as a sensitive and specific immunohistochemical marker, while its expression is also essential for the maintenance of Schwann cell and melanocyte lineages throughout adulthood.

Flow cytometric analysis of SOX10 expression on 4% PFA fixed 90% methanol permeabilized A375 cells. Cells from the A375 (Human malignant melanoma epithelial cell, Right) or HeLa (Human cervix adenocarcinoma epithelial cell, Left) was stained with either Alexa Fluor® 647 Rabbit IgG Isotype Control (Black line histogram) or SDT SOX10 Recombinant Rabbit mAb (Alexa Fluor® 647 Conjugate) (Red line histogram) at 1/2000 dilution (0.1 μg), cells without incubation with primary antibody and secondary antibody (Blue line histogram) was used as unlabelled control. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Immunocytochemistry

ICC shows positive staining in A375 cells (top panel) and negative staining in HeLa cells (below panel). Anti-SOX10 Recombinant Rabbit mAb (Alexa Fluor® 647 Conjugate) antibody was used at 1/200 dilution (meganta) and incubated overnight at 4°C. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue).

Immunofluorescence

IF shows positive staining in paraffin-embedded human melanoma. Anti-SOX10 Recombinant Rabbit mAb (Alexa Fluor® 647 Conjugate) antibody was used at 1/200 dilution (meganta) and incubated overnight at 4°C. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.

Anti-SOX10 Recombinant Rabbit mAb (Alexa Fluor® 647 Conjugate) antibody was used at 1/200 dilution (meganta) and incubated overnight at 4°C. Counterstained with DAPI (Blue).
