Background
SEK1/MKK4 (also known as MAP2K4, MEK4, JNKK1) is a 399-amino-acid dual-specificity protein kinase located on chromosome 17p11.2 that functions as the immediate upstream activator of both JNK and p38 MAPKs in response to diverse extracellular stresses such as osmotic shock, UV radiation, and cytokines; after phosphorylation by upstream MAP3Ks like MEKK1/2, SEK1/MKK4 phosphorylates JNK and p38 on their Thr-Pro-Tyr motifs, thereby linking stress signals to transcriptional and apoptotic programs, and its activity and stability are subjected to negative feedback control through JNK-mediated phosphorylation of the E3 ubiquitin ligase Itch, which then ubiquitinates and targets SEK1/MKK4 for proteasomal degradation .

IHC shows positive staining in paraffin-embedded human skeletal muscle. Anti-SEK1/MKK4 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
