Background

DAZL (Deleted in Azoospermia-Like) is a highly conserved, germ-cell-specific RNA-binding protein that orchestrates key post-transcriptional events across vertebrate gametogenesis. It carries an N-terminal RNA-recognition motif and a C-terminal DAZ repeat that jointly bind U-rich sequences in the 3′ UTRs of target mRNAs, enabling DAZL to recruit poly(A)-binding proteins (PABPs) and stimulate cap-dependent translation initiation independently of the poly(A) tail length. Expressed from primordial germ cell (PGC) specification through meiotic progression in both sexes, DAZL licenses these cells to respond to retinoic acid, thereby promoting meiotic entry, synaptonemal complex formation via SYCP3, and spindle assembly during oocyte maturation. In vivo, loss of DAZL causes complete germ-cell depletion and infertility, whereas haploinsufficiency yields abnormal sperm and ovarian insufficiency, underscoring its role as a master regulator of germline survival and differentiation.

IHC shows positive staining in paraffin-embedded mouse testis. Anti-DAZL antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Negative control: IHC shows negative staining in paraffin-embedded mouse cerebral cortex. Anti-DAZL antibody was used at 1/2000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin.
