Background

Phospho-p38 MAPK (Thr180) is the dual-phosphorylated, enzymatically active form of the p38 mitogen-activated protein kinase in which the conserved threonine residue at position 180 (within the canonical Thr-X-Tyr activation motif, TGY in human p38α) is phosphorylated, together with Tyr182, by upstream MAP kinase kinases (MKK3/6 or MKK4), resulting in a conformational switch that opens the ATP- and substrate-binding sites, thereby enabling the kinase to transduce stress, inflammatory, and cytokine signals to downstream transcription factors (e.g., ATF-2, CHOP, MEF2C) and other effectors, ultimately governing cellular responses such as apoptosis, differentiation, and cytokine production.
