Background
Phospho-CrkL (Tyr207), the tyrosine-207-phosphorylated form of the CRK-like adaptor protein CrkL, is a 34 kDa signaling scaffold that contains SH2 and SH3 domains enabling it to bind multiple partners and propagate signals through MAPK, PI3K and STAT cascades; phosphorylation at Tyr207 (the CrkL counterpart of Tyr221 in CrkII) is a key regulatory switch controlling cytoskeletal reorganization, immune-cell signaling, cell proliferation and migration, and its dysregulation contributes to Philadelphia-chromosome-positive leukemias via constitutive activation with BCR-ABL, while developmental perturbations are linked to congenital heart defects, making p-CrkL(Tyr207) a focal point in oncogenic and developmental research .
