Background
The NUT protein, which stands for nuclear protein in testis, is a transcription factor that plays a crucial role in embryonic development. It is typically found in the testes and is involved in the regulation of gene expression. However, when it is abnormally expressed or translocated in other tissues, it can lead to the development of certain types of cancers, particularly NUT midline carcinoma (NMC), a rare but aggressive malignancy. The presence of the NUT protein in cancer cells is often associated with poor prognosis, and it has become an important target for research in cancer diagnostics and therapeutics.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human testis. Anti-NUT antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
