Background
MUC5B, encoded on chromosome 11p15.5, is a 5,700-amino-acid gel-forming mucin produced chiefly by mucosal epithelial cells in the respiratory tract, gastrointestinal tract, cervix, and salivary glands; it polymerizes via N-terminal von Willebrand factor-like D1-D2-D′-D3 domains that disulfide-link into massive, highly glycosylated (O-linked) linear chains whose extensive carbohydrate content (up to 80 % of mass) endows viscoelastic, lubricating, and antimicrobial barrier properties essential for mucociliary clearance and host defense.
