Background

Merlin protein, also known as neurofibromin 2 (NF2), is a crucial tumor suppressor protein encoded by the NF2 gene. It plays a vital role in regulating cell proliferation, differentiation, and apoptosis. Merlin acts as a negative regulator of the Hippo signaling pathway, which is essential for controlling organ size and cell fate determination. When functioning normally, Merlin helps to inhibit the overgrowth of cells and prevents the formation of tumors. Mutations in the NF2 gene that lead to a loss of functional Merlin protein are associated with the development of various types of cancer, including schwannomas, meningiomas, and mesotheliomas. In addition to its role in cancer, Merlin is also involved in the regulation of cell adhesion and the maintenance of cellular architecture. Ongoing research is focused on understanding the detailed mechanisms of Merlin's action and exploring potential therapeutic strategies targeting the Merlin-Hippo pathway for cancer treatment.
