Background
M-Cadherin (Cadherin-15), a classical calcium-dependent cell-adhesion glycoprotein, is primarily expressed in developing skeletal muscle, satellite cells and cerebellum, where it mediates homophilic binding to organize myoblast fusion, muscle fiber formation and neuronal layer integrity by assembling adherens junctions through catenin-dependent linkage to the actin cytoskeleton ; despite its name, germline deletion in mice is largely compensated by N-cadherin, yet human mutations causing loss of adhesive function are linked to autosomal-dominant mental retardation-3, underscoring a non-redundant role in synaptic and cognitive development .

Immunocytochemistry
ICC shows positive staining in C2C12 cells (top panel) and negative staining in NIH/3T3 cells (below panel). Anti- M-cadherin/cadherin-15 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
