Background
Low molecular weight protein tyrosine phosphatase (LMPTP), an 18 kDa cytosolic class II PTP encoded by the ACP1 gene, exists as two splice isoforms (LMPTP-A and LMPTP-B), is ubiquitously expressed with especially high levels in adipocytes, and functions as a negative regulator of insulin signalling by dephosphorylating the insulin receptor and its substrates; human genetic studies show that low-activity ACP1 alleles are associated with reduced glycaemia, while experimental knockdown or genetic deletion of LMPTP in obese mice improves glucose tolerance and insulin sensitivity—mainly via hepatic effects—demonstrating that LMPTP drives obesity-linked insulin resistance and type 2 diabetes, thus representing an attractive drug target for metabolic disorders.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human breast cancer. Anti-LMPTP antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in HEK293 cells. Anti-LMPTP antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
