Background
JNK1, JNK2, and JNK3 are the three main isoforms of the c-Jun N-terminal Kinase family, which belongs to the MAPK (Mitogen-Activated Protein Kinase) superfamily. They are critical signaling molecules that translate extracellular stimuli into intracellular responses. Encoded by distinct genes (MAPK8, MAPK9, and MAPK10, respectively), these three isoforms share high structural similarity but also exhibit significant functional differences: JNK1 and JNK2 are ubiquitously expressed across various tissues and are involved in regulating core cellular processes in response to stresses such as oxidative stress and genotoxic stress, as well as inflammatory cytokines. These processes include cell proliferation, differentiation, apoptosis, and metabolic homeostasis. In contrast, the expression of JNK3 is primarily restricted to the brain, heart, and testes, with particularly high abundance in neurons. This localized expression assigns JNK3 a central role in central nervous system-specific functions, such as regulating neuronal apoptosis and synaptic plasticity, and it is closely implicated in the pathology of various neurodegenerative diseases like Alzheimer's and Parkinson's. Although they share common substrates, such as the transcription factor c-Jun (which they activate by phosphorylating its N-terminus), subtle differences in their promoter activity, binding preferences for scaffold proteins, and subcellular localization often lead to unique, and sometimes opposing, functions in specific contextual settings. This intricate interplay forms a sophisticated signaling network that plays a vital role in determining cell fate and the organism's response to stress.

Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti- JNK1 + JNK2 + JNK3 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
