Background
Interleukin-1 beta (IL-1β) is a pro-inflammatory cytokine that plays a crucial role in the immune response. It is produced by various cell types, including macrophages and monocytes, in response to infections or tissue injury. IL-1β helps to initiate and amplify the inflammatory response by promoting the expression of other inflammatory mediators and recruiting immune cells to the site of infection or damage. It also has effects on a wide range of physiological processes, such as fever induction, modulation of the hypothalamic-pituitary-adrenal axis, and regulation of metabolism. Dysregulation of IL-1β has been implicated in numerous chronic inflammatory diseases, like rheumatoid arthritis and inflammatory bowel disease, making it an important target for therapeutic interventions.

Immunocytochemistry
ICC analysis of THP-1 cells treated with TPA (80 nM,16h), then add LPS (100 ng/ml,6h), then add BFA (300 ng/ml, 3h) (top panel) and untreated THP-1 cells (below panel). Anti- IL-1 beta antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
