Background

14-3-3 protein sigma (SFN), also known as Stratifin, is a crucial member of the 14-3-3 family. It functions as a phospho-serine/phospho-threonine binding protein that regulates a wide array of cellular processes, including cell cycle progression, apoptosis, and stress response. A key and well-characterized role of 14-3-3 sigma is its involvement in the DNA damage checkpoint, where it is transcriptionally activated by p53 and helps arrest the cell cycle. It often acts as a tumor suppressor, and its expression is frequently lost in various cancers through epigenetic silencing.
