Background
HADHA, which stands for hydroxyacyl-CoA dehydrogenase trifunctional multienzyme complex alpha subunit, is a crucial protein involved in the mitochondrial beta-oxidation of fatty acids. It is a part of the trifunctional protein complex that catalyzes three sequential steps in the breakdown of long-chain fatty acids into acetyl-CoA, a key molecule in cellular energy production. This protein is essential for the proper metabolism of fats and is particularly important in tissues with high energy demands such as the liver and heart. Mutations in the HADHA gene can lead to severe metabolic disorders, including long-chain 3-hydroxyacyl-CoA dehydrogenase deficiency, which can cause symptoms ranging from hypoglycemia and cardiomyopathy to more severe neurological complications, highlighting its critical role in maintaining metabolic homeostasis.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human cardiac muscle. Anti-HADHA antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti- HADHA antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
