Background
Gli2 protein is a crucial transcription factor in the Hedgehog signaling pathway, playing a significant role in embryonic development and tissue homeostasis. It is involved in regulating cell proliferation, differentiation, and survival in various tissues. Gli2 acts as both an activator and repressor of gene expression, depending on the context and the presence of Hedgehog ligands. In the absence of signaling, Gli2 is processed into a repressor form that inhibits target gene transcription. However, when Hedgehog ligands bind to their receptors, Gli2 is converted into an activator form that promotes the expression of genes essential for proper development and tissue repair. Dysregulation of Gli2 activity has been implicated in various diseases, including cancer and developmental disorders, making it an important target for therapeutic interventions.

Immunocytochemistry
ICC shows positive staining in 293T cells. Anti-Gli2 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
