Background
GABARAP (GABA type A receptor-associated protein) is a 14-kDa cytosolic mammalian ATG8-family protein that, upon activation by ATG7 and ATG3, covalently conjugates to phosphatidylethanolamine on forming autophagosomes, thereby acting as a pivotal scaffold for recruitment and tethering of autophagy receptors such as LC3-interacting region (LIR)-containing cargo adaptors, as well as for ULK1 complex and PI3-kinase complex I assembly; beyond autophagy, it participates in intracellular trafficking, receptor stabilization, cytoskeletal reorganization, and innate immunity by modulating GABAA receptor clustering at synapses, tethering ER to the Golgi via golgin-160, and interacting with NDP52/TBK1 to restrict cytosolic bacteria, and its dysregulation is implicated in neurodegeneration, cancer, and infectious diseases.
