Background
Fbx32, also known as Atrogin-1 or MAFbx, is an F-box protein that functions as the substrate-recognition component of the Skp1-Cul1-F-box (SCF) E3 ubiquitin ligase complex, selectively targeting key muscle proteins such as MyoD, eIF3-f, and calcineurin for polyubiquitination and proteasomal degradation, thereby regulating skeletal muscle atrophy in response to catabolic signals like glucocorticoids, inflammatory cytokines, and denervation, while its expression is controlled by the FoxO transcription factors and its inhibition or deletion attenuates muscle wasting in various disease models.
