Background
Cortactin is an actin-binding scaffold protein encoded by the human CTTN gene that localizes to membrane ruffles, lamellipodia, invadopodia and other actin-rich structures, where it stabilizes Arp2/3-nucleated branched actin networks through its N-terminal acidic (NTA) domain and tandem repeat region, while its C-terminal SH3 domain recruits multiple binding partners including N-WASP, dynamin-2, and cortactin-binding protein-2 to coordinate cytoskeletal remodeling, endocytosis, cell migration, and extracellular matrix degradation; overexpression or phosphorylation of cortactin—especially at tyrosine residues 421, 466, and 482 by Src family kinases—amplifies these activities, driving tumor cell invasion, metastasis, and resistance to anoikis, and its gene is frequently amplified and its protein overexpressed in a wide range of epithelial cancers, making cortactin both a biomarker of poor prognosis and a potential therapeutic target.

Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti- Cortactin antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
