Background
CNOT1 (CCR4-NOT transcription complex subunit 1) is a crucial protein encoded by the CNOT1 gene and serves as the central scaffold for the CCR4-NOT complex, which is a master regulator of gene expression involved in mRNA deadenylation, transcriptional regulation, and protein ubiquitination. This complex plays a significant role in various cellular processes, including mRNA degradation, DNA repair, and circadian rhythm regulation. CNOT1 itself does not possess catalytic activity but is essential for maintaining the integrity of the CCR4-NOT complex and binding to other subunits and RNA-binding proteins. It is also implicated in neurodevelopmental disorders, as de novo variants in CNOT1 have been associated with intellectual disability, seizures, and other developmental issues.

Secondary antibody: Goat Anti-rabbit IgG, (H+L), HRP conjugated at 1/10000 dilution

Immunocytochemistry
ICC shows positive staining in 293T cells. Anti-CNOT1 antibody was used at 1/50 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
