Background
CDC7 (cell division cycle 7-related protein kinase) is a highly conserved, nuclear serine/threonine kinase encoded by the CDC7 gene that governs the initiation of DNA replication at the G1/S transition by forming an active complex with its regulatory subunit Dbf4 (ASK in mammals); this CDC7-Dbf4/ASK complex phosphorylates the MCM2–7 helicase components of the pre-replication complex, thereby triggering origin firing and S-phase entry, while its activity is kept constant throughout the cell cycle by post-translational control rather than changes in CDC7 levels. Loss or inhibition of CDC7 arrests DNA synthesis and can lead to p53-mediated apoptosis, whereas its overexpression—frequently observed in colorectal, lung, breast and hematological malignancies—correlates with neoplastic transformation and may be exploited therapeutically with CDC7 inhibitors.

Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti- CDC7 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
