Flow cytometric analysis of IL-17A expression in Th17 polarized C57BL/6 mouse splenocytes. Th17 polarized C57BL/6 mouse splenocytes were harvested and fixed with 4% PFA and permeabilized with Intracellular Fixation & Permeabilization Buffer Set. The cells were then stained with FITC Rat anti-Mouse CD4 antibody (S0B1646) and either Alexa Fluor® 647 Rat IgG1, κ Isotype Control or SDT Alexa Fluor® 647 Rat Anti-Mouse IL-17A Antibody at 1.25μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
IL-17A protein a is key cytokine produced by various immune cells such as Th17 cells. It plays a crucial role in the immune response by promoting inflammation. It can induce the expression of various pro-inflammatory molecules in different cell types, including chemokines that recruit neutrophils and other immune cells to the site of infection or injury. IL-17A is involved in the defense against certain pathogens, but its excessive production is also associated with the pathogenesis of several autoimmune and inflammatory diseases like psoriasis and rheumatoid arthritis. Targeting IL-17A has become an important therapeutic strategy in the treatment of these conditions.
