Background

Protein tyrosine phosphatase 1B (PTP1B), encoded by the PTPN1 gene, is a 50 kDa non-receptor intracellular tyrosine phosphatase anchored to the cytoplasmic face of the endoplasmic reticulum that acts as a key negative regulator of both the insulin and leptin signaling cascades by dephosphorylating the insulin receptor, IRS-1, JAK2, and leptin receptor, thereby dampening downstream PI3K/AKT and STAT3 signaling; PTP1B-null mice are lean, insulin-hypersensitive, and resistant to diet-induced obesity, while elevated PTP1B expression is associated with insulin resistance, type 2 diabetes, obesity, and several cancers (especially breast and ovarian cancer), and selective PTP1B inhibitors are actively explored as therapeutics for metabolic and oncologic disorders.
