Background
Protein kinase C theta (PKC-θ), encoded by the PRKCQ gene on chromosome 10p15.1, is a calcium-independent, phospholipid-dependent serine/threonine kinase selectively expressed in hematopoietic cells—especially platelets and T lymphocytes—where it transduces T-cell receptor signals to activate NF-κB and AP-1, thereby orchestrating T-cell activation, survival, and distinct Th17-mediated immune responses, while also promoting oncogenic growth and anoikis resistance in triple-negative breast cancer cells, and its kinase activity is considered a potential therapeutic target in autoimmunity and cancer.
