Background
Alpha smooth muscle actin (alpha-SMA, acetyl E3) and ACTG2 (actin gamma 2, acetyl E3) are two important actin isoforms primarily involved in smooth muscle cell contraction and cytoskeletal maintenance. Alpha-SMA is a key component of the contractile apparatus in muscle tissues, particularly abundant in vascular, gastrointestinal, and urinary tract smooth muscles. Its acetylation modification (acetyl E3) may influence its function or stability. ACTG2 (γ2-actin) is a smooth muscle-specific γ-actin predominantly expressed in visceral smooth muscles, such as those in the intestines, and its acetylation (acetyl E3) also plays a role in cell motility and structural support. Studies indicate that both proteins can be detected by specific antibodies (e.g., Abcam's [E184] antibody) in immunohistochemistry, with α-SMA showing potential in inhibiting scar formation in fibroblast contraction assays. Their genes and protein structures are highly conserved, and they functionally cooperate in regulating mechanical contraction and cellular morphology in smooth muscle.

Immunohistochemistry

Anti-alpha smooth muscle Actin (acetyl E3) + ACTG2 (acetyl E3) antibody was used at 1/4000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
