Background
Human IL-4R (Interleukin-4 Receptor) is a critical cell surface receptor involved in immune regulation and inflammatory responses, composed of the IL-4Rα chain (CD124) paired with either the common γ-chain (γc, CD132) or IL-13Rα1 to form Type I or Type II receptors. It binds cytokines IL-4 and IL-13, activating JAK-STAT signaling pathways that modulate immune cell functions, particularly in hematopoietic cells such as B cells, T cells, macrophages, and mast cells. IL-4R signaling promotes Th2 cell differentiation, IgE antibody class switching, and anti-inflammatory responses, playing a central role in allergic and immune-mediated diseases. Dysregulation of IL-4R is associated with conditions like asthma, atopic dermatitis, and autoimmune disorders. Due to its therapeutic potential, IL-4R has been successfully targeted by drugs such as dupilumab, offering effective treatment for allergic diseases. Ongoing research continues to explore IL-4R's broader roles in immunity and inflammation, paving the way for novel therapies targeting immune dysregulation.

Linearity
The concentrations of Human IL-4R were measured and interpolated from the target standard curves and corrected for sample dilution.
Sample is undiluted samples are as follows: human plasma (25%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 565.71 pg/mL in human plasma #1. The mean target concentration was determined to be 739.72 pg/mL in human plasma #2.
Linearity
The concentrations of Human IL-4R were measured and interpolated from the target standard curves and corrected for sample dilution.
Sample is undiluted samples are as follows: human serum (25%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 464.86 pg/mL in human serum #1. The mean target concentration was determined to be 472.25 pg/mL in human serum #2.
Demonstration of specificity in mice
The sample dilution factor was 4-fold.
The added human IL-4R was detected in mouse serum, with a recovery rate of 101%.
