Background
TREM2 (Triggering Receptor Expressed on Myeloid Cells 2) is a ~40 kDa single-pass type I transmembrane glycoprotein of the immunoglobulin superfamily that is almost exclusively expressed by microglia in the brain, where together with its signaling adaptor DAP12 it forms a receptor complex that upon binding anionic lipids, apolipoprotein E, or aggregated β-amyloid initiates phosphorylation cascades that regulate microglial survival, proliferation, metabolic reprogramming toward glycolysis and oxidative phosphorylation, and phagocytosis of apoptotic neurons and extracellular plaques, thereby maintaining central nervous system homeostasis; genetic loss-of-function variants strongly increase the risk of Alzheimer’s disease, and its ectodomain is proteolytically cleaved by ADAM10 and ADAM17 to release a soluble fragment (sTREM2) that is elevated in cerebrospinal fluid in early AD and can itself act as an immunomodulatory ligand amplifying microglial responses to pathology.

Immunocytochemistry
ICC shows positive staining in J774A.1 cells (top panel) and negative staining in Neuro-2a cells (below panel). Anti-Trem2 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
