Background
Transforming Growth Factor-beta 1 (TGF-β1) is a multifunctional 25 kDa homodimeric cytokine that belongs to the TGF-β superfamily and is secreted by virtually all somatic cells as a large latent complex bound to latency-associated peptide and latent TGF-β binding protein; upon activation by proteases or integrins it signals through a heteromeric receptor complex of TβRII and ALK5 (TβRI) serine/threonine kinases to phosphorylate Smad2/3, which then complex with Smad4 to regulate transcription of hundreds of genes that orchestrate pleiotropic effects ranging from embryogenesis, extracellular matrix deposition, immune suppression and wound healing to cell cycle arrest, apoptosis, and epithelial-mesenchymal transition, with dysregulation implicated in fibrosis, cancer, and autoimmune disease.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human bone marrow. Anti-TGF beta 1 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
