Background
Stat5 (Signal Transducer and Activator of Transcription 5) is a latent cytoplasmic transcription factor that becomes phosphorylated on a conserved tyrosine (Tyr694 in human STAT5a) by activated Janus kinases—primarily JAK2—following cytokine (e.g., IL-2, IL-3, GM-CSF, GH, PRL, EPO, TPO) or oncogenic tyrosine-kinase receptor (e.g., BCR-ABL, FLT3-ITD, JAK2V617F) signaling, whereupon it forms parallel homo- or heterodimers, translocates into the nucleus, binds the consensus DNA motif TTCNNNGAA within enhancers/promoters of target genes like CISH, BCL2, MYC, CCND1, PIM1 and SOCS proteins, and thereby orchestrates proliferation, survival, self-renewal and differentiation programs in hematopoietic, mammary and other cell types; persistent Stat5 activation is a driver of myeloproliferative neoplasms, acute leukemias and breast cancers, making its upstream kinases and the Stat5 SH2 or DNA-binding domains attractive therapeutic targets.
