Background
NUP98 (nucleoporin 98) is a 98-kDa component of the nuclear pore complex (NPC) generated by autoproteolytic cleavage of a 186-kDa precursor that also yields NUP96 . Anchored on the nucleoplasmic face of the NPC, it contains N-terminal GLFG/FG repeats that serve as docking sites for transport receptors during nucleocytoplasmic trafficking of proteins and RNAs, while its C-terminal RNA-binding domain participates in mRNA export and gene-expression regulation . Beyond transport, NUP98 dynamically associates with mitotic structures to control spindle assembly and mitotic progression, forms liquid-droplet compartments that sequester damaged DNA to safeguard genome integrity, and acts as a transcriptional coactivator or corepressor by localizing to developmental gene loci. Chromosomal translocations that fuse the N-terminal portion of NUP98 to over 30 partner proteins (e.g., homeodomain transcription factors) generate potent oncoproteins that drive aggressive forms of acute myeloid leukemia and T-cell acute lymphoblastic leukemia, highlighting its pivotal role in both normal cellular physiology and oncogenesis.
