Background
NFAT1 (Nuclear Factor of Activated T-cells 1) is a ~115 kDa transcription factor of the NFAT family that, upon cooperative binding of Ca²⁺/calmodulin-activated phosphatase calcineurin to its conserved PxIxIT and LxVP motifs, undergoes rapid dephosphorylation at multiple serine-rich motifs within its regulatory domain, exposing a nuclear localization sequence that drives its translocation from cytosol to nucleus where it cooperates with Fos/Jun, GATA, and IRF transcription partners to bind the consensus DNA element 5’-A/GAAGGAAA-3’ and activate transcription of cytokine genes (IL-2, IL-4, TNF-α), surface receptors (CD40L), and developmental regulators, while its activity is terminated by glycogen synthase kinase-3β (GSK-3β) and casein kinase-1 (CK1)-mediated re-phosphorylation followed by Crm1-dependent nuclear export; dysregulated NFAT1 signaling contributes to cardiac hypertrophy, autoimmune pathologies, and neoplastic growth, making calcineurin–NFAT1 interaction a pharmacological target of immunosuppressive drugs such as cyclosporine A and tacrolimus.
