Background
The insulin receptor β-subunit is the 95 kDa transmembrane tyrosine kinase moiety of the heterotetrameric insulin receptor, spanning the plasma membrane once with its short extracellular domain stabilizing disulfide linkage to the α-subunit and its large cytoplasmic tail harboring an ATP-binding site and catalytic domain that, upon insulin-induced autophosphorylation of specific tyrosines such as Y1158, Y1162 and Y1163, recruits and phosphorylates IRS and Shc adaptor proteins to initiate PI3K-Akt and MAPK signaling cascades governing glucose uptake, glycogen synthesis, lipogenesis and cell growth, while mutations in the β-subunit that impair kinase activity or endocytosis are linked to severe insulin resistance syndromes including Rabson–Mendenhall and type A diabetes.
