Background
The GCLC (glutamate–cysteine ligase catalytic subunit) protein is the catalytic subunit of glutamate cysteine ligase (GCL, formerly γ-glutamylcysteine synthetase), the enzyme that catalyzes the first, rate-limiting step in glutathione (GSH) synthesis by forming γ-glutamylcysteine from glutamate and cysteine using ATP; this ~73 kDa subunit contains all substrate and cofactor binding sites necessary for catalysis, while the regulatory GCLM subunit increases its catalytic efficiency when complexed in the holoenzyme, and mutations in GCLC are associated with hemolytic anemia due to GSH deficiency and susceptibility to myocardial infarction [^5].
