Background

Choline acetyltransferase (ChAT) is a 69-kDa cytosolic transferase enzyme encoded by the CHAT gene that catalyzes the single-step acetylation of choline with acetyl-CoA to synthesize the neurotransmitter acetylcholine (ACh) in the presynaptic terminals of cholinergic neurons throughout the central and peripheral nervous systems; it is synthesized in the soma, trafficked to nerve endings via axoplasmic flow, exists in soluble and membrane-bound pools, displays alternative splicing that can generate larger nuclear variants, and is considered the most specific phenotypic marker for cholinergic neurons whose functional integrity underlies memory, cognition, arousal, and motor control, while its loss or dysfunction is closely linked to the cholinergic deficits seen in Alzheimer’s disease, amyotrophic lateral sclerosis, schizophrenia, and other neurodegenerative or neuropsychiatric disorders .

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-Choline Acetyltransferase/ChAT antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
