Background
CD42a, also known as glycoprotein IX (GPIX), is a small, single-pass transmembrane glycoprotein expressed on platelets and megakaryocytes that non-covalently associates with glycoproteins Ibα, Ibβ, and V to form the GPIb-V-IX complex, which acts as the platelet receptor for von Willebrand factor (vWF), enabling vWF-dependent platelet adhesion to injured vessel walls and initiating arterial hemostasis; CD42a contains leucine-rich repeats, is encoded by GP9 on chromosome 3q21.3, and mutations in this gene cause Bernard–Soulier syndrome, a bleeding disorder characterized by giant platelets and impaired aggregation.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human spleen. Anti-CD42a antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
Negative control: IHC shows negative staining in paraffin-embedded human cerebral cortex. Anti-CD42a antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
