Background

Caspase-1, the canonical member of the inflammatory caspase subfamily, is synthesized as a 45 kDa zymogen that assembles into the ~700 kDa inflammasome platform via homotypic CARD–CARD interactions with ASC and NLRP proteins; proximity-induced dimerization and trans-autoproteolysis generate the active p20/p10 tetramer whose p10 subunit forms a heterodimeric active site that cleaves the tetrapeptide motifs WEHD and YVAD, liberating the pro-inflammatory cytokines IL-1β and IL-18, triggering pyroptosis through gasdermin-D pore formation, and thereby orchestrating innate immune defense, metabolic regulation, and, when dysregulated, autoinflammatory diseases such as CAPS, atherosclerosis, and Alzheimer’s pathology.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded mouse colon. Anti- Caspase-1 antibody was used at 1/250 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
