Background
Myelin protein zero (P0, MPZ) is a 28–30 kDa type I transmembrane glycoprotein that constitutes 50–70 % of total protein in peripheral nervous system (PNS) compact myelin, where it is expressed continuously by myelinating Schwann cells until mature myelin is formed; its extracellular immunoglobulin-like domain mediates homophilic adhesion between apposing membrane pairs to generate the characteristic intraperiod line, while its short cytoplasmic tail cooperates with myelin basic protein to zipper the cytoplasmic faces, enabling the tight membrane stacking that gives PNS myelin its insulating properties, and mutations in the MPZ gene underlie a spectrum of incurable demyelinating peripheral neuropathies such as Charcot–Marie–Tooth disease by disrupting these adhesive and structural functions.

Anti-Myelin protein zero antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunofluorescence
IF shows positive staining in paraffin-embedded human sciatic nerve of foot. Anti-Myelin protein zero antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
