Background
HPV18 E7 is a ~100-amino-acid, zinc-binding, dimeric oncoprotein encoded early in the HPV18 life cycle; via its conserved CR1/CR2 motifs it binds the retinoblastoma tumor suppressor pRb and related pocket proteins, displacing E2F transcription factors and forcing the infected epithelial cell into S-phase, while additional interactions with APC/C-Cdh1, p21Cip1 and p27Kip1 override checkpoint control, promote centrosome duplication errors and genomic instability, and together with its C-terminal CXXC motifs that coordinate Zn2+ for dimerization, ensure sustained proliferation and the oncogenic progression that underlies the majority of HPV18-driven cervical and other anogenital cancers.
