Background

CD133, also known as Prominin-1, is a pentaspan membrane glycoprotein expressed on the surface of various stem cells and cancer stem cells (CSCs). It plays a crucial role in tumorigenesis, metastasis, recurrence, and drug resistance, and is involved in the regulation of cellular energy metabolism, affecting glucose uptake, autophagy, and ATP synthesis. Moreover, the function of CD133 is not limited to cancer cells; it is also expressed in differentiated cells and stem/progenitor cells in normal tissues and is involved in multiple cellular processes, coordinating signaling pathways that are often deregulated in cancer, such as the PI3K/Akt signaling and Wnt/β-catenin pathways.

IP

CD133 Rabbit mAb at 1/50 dilution (1 µg) immunoprecipitating CD133 in 0.4 mg HT-29 whole cell lysate.
Western blot was performed on the immunoprecipitate using CD133 Rabbit mAb at 1/1000 dilution.
Secondary antibody (HRP) for IP was used at 1/1000 dilution.
Lane 1: HT-29 whole cell lysate 5 µg (Input)

Lane 3: Rabbit monoclonal IgG IP in HT-29 whole cell lysate
Predicted MW: 97 kDa
Observed MW: 110~190 kDa

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human kidney. Anti-CD133 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

IHC shows positive staining in paraffin-embedded human pancreas.

IHC shows positive staining in paraffin-embedded human endometrial cancer.
