Background
Human Thyroid Stimulating Hormone (TSH), a glycoprotein hormone secreted by the anterior pituitary gland, plays a pivotal role in regulating thyroid function by stimulating the synthesis and secretion of thyroid hormones (T3 and T4), which are essential for metabolism, growth, and development. TSH binds to TSH receptors on thyroid follicular cells and its secretion is regulated by a negative feedback loop involving thyrotropin-releasing hormone (TRH) from the hypothalamus and circulating T3/T4 levels. As the most sensitive marker for thyroid disorders, elevated TSH indicates hypothyroidism while suppressed TSH suggests hyperthyroidism. With a molecular weight of ~28 kDa consisting of α- and β-subunits, TSH is typically measured using immunoassays (ELISA, CLIA, RIA) with a normal range of 0.4-4.0 mIU/L. Clinically, TSH testing is indispensable for diagnosing various thyroid conditions (Hashimoto's thyroiditis, Graves' disease), monitoring levothyroxine therapy, and assessing thyroid function during pregnancy, making it the cornerstone of thyroid function evaluation due to its high diagnostic reliability.

Linearity
The concentrations of TSH were measured and interpolated from the target standard curves and corrected for sample dilution.
The sample is undiluted samples are as follows: Human serum was 16fold dilution. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 187.67 pg/mL in human serum.

To assess the linearity of the assay, two samples were spiked with high concentrations of Human TSH in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
