Background
Human Insulin-like Growth Factor-1 (IGF-1) is a 7.6 kDa polypeptide hormone structurally similar to insulin, playing a crucial role in growth, development, and metabolic regulation. Primarily produced in the liver under stimulation by growth hormone (GH), IGF-1 mediates many of GH's anabolic and mitogenic effects via activation of the IGF-1 receptor (IGF-1R), a tyrosine kinase receptor that triggers downstream signaling pathways such as PI3K/AKT and MAPK/ERK. IGF-1 is essential for: Growth & Development: Promotes cellular proliferation, differentiation, and tissue growth, particularly in bone, muscle, and nervous systems. Metabolic Regulation: Enhances glucose uptake, protein synthesis, and inhibits apoptosis. Tissue Repair: Supports regeneration in muscle, cartilage, and neuronal tissues. Clinically, recombinant human IGF-1 (rhIGF-1, e.g., mecasermin) is used to treat growth disorders like Laron syndrome and severe primary IGF-1 deficiency. It is also investigated for conditions such as diabetes, neurodegenerative diseases, and muscle wasting disorders. However, its dysregulation is associated with cancer progression and acromegaly.
