Background
Human Interferon Beta (IFNβ) is a naturally occurring cytokine belonging to the type I interferon family, primarily produced by fibroblasts and immune cells in response to viral infections or other inflammatory stimuli. It plays a critical role in the innate immune response by binding to the IFNα/β receptor (IFNAR), activating the JAK-STAT signaling pathway, and inducing the expression of interferon-stimulated genes (ISGs). These genes exert broad antiviral, immunomodulatory, and antiproliferative effects. IFNβ has been widely utilized as a therapeutic agent, particularly in the treatment of relapsing-remitting multiple sclerosis (MS), where it helps reduce inflammation and modulate immune activity. Recombinant forms of IFNβ (e.g., IFNβ-1a and IFNβ-1b) are clinically approved and demonstrate efficacy in decreasing relapse rates and slowing disease progression. Key functions of IFNβ include: Antiviral Activity: Enhances cellular resistance to viral infections. Immunoregulation: Suppresses pro-inflammatory cytokines (e.g., TNFα, IL-12) and promotes anti-inflammatory responses. Antiproliferative Effects: Inhibits the growth of certain tumor cells.

The concentrations of Human IFNβ were measured and interpolated from the target standard curves and corrected for sample dilution.
Sample is undiluted samples are as follows: human PBMC cells stimulated with 2ug/ml polyI:C for 24 hours (100%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 14.35 pg/mL in stimulated human PBMC supernatant.
Linearity
The concentrations of Human IFNβ were measured and interpolated from the target standard curves and corrected for sample dilution.
Sample is undiluted samples are as follows: A549 cells stimulated with 2ug/ml polyI:C for 24 hours (100%). The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 527.76 pg/mL in stimulated A549 supernatant.
To assess the linearity of the assay, sample was spiked with high concentrations of Human IFNβ in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
