Background
Mouse Klotho (mKL) is a 1014-aa type-I trans-membrane glycoprotein of the glycosidase-1 super-family that is expressed mainly in distal convoluted tubule cells of the kidney, choroid plexus of the brain, and adrenal zona glomerulosa. A soluble 130-kDa form is generated by ADAM10/17 proteolytic shedding and circulates in plasma and CSF. Klotho functions as an obligate co-receptor for FGF23, thereby regulating phosphate and vitamin-D homeostasis; It also exhibits intrinsic β-glucuronidase activity that activates TRPV5/6 channels for renal Ca²⁺ reabsorption. Homozygous Klotho-deficient (kl/kl) mice on a mixed C57BL/6J, FVB, and C3H/J background develop a premature-aging phenotype—growth retardation, osteoporosis, arteriosclerosis, skin atrophy, emphysema, infertility, and death by 8–9 weeks. Conversely, transgenic over-expression extends lifespan and improves cognition, synaptic plasticity, and stress resilience.
